Reconstitution
What it means to reconstitute a lyophilized vial, why the diluent choice matters, and how the process fits into the arithmetic of concentration.

Overview
Reconstitution is the step in which a fixed volume of a sterile diluent is added to a lyophilized vial so that the dry peptide dissolves into solution. Once reconstituted, the solution can be drawn into a syringe and its volume can be measured.
The peptide mass inside the vial does not change during reconstitution. Only the total volume the material occupies changes. That is why concentration — mass ÷ volume — is fully determined by what mass was in the vial and what volume of diluent you added.
Why it matters
Every measurement downstream depends on reconstitution being done correctly. If the diluent volume is wrong, the concentration is wrong, and every syringe reading derived from that concentration is wrong.
Key concepts
Slow, targeted addition
The diluent is typically added slowly and directed at the vial wall rather than sprayed onto the cake. This reduces mechanical stress on the peptide and reduces bubbling.
Swirl, do not shake
Vigorous shaking can denature peptides and generate foam that traps material. Gentle swirling encourages dissolution without mechanical damage.
Full dissolution
A properly reconstituted vial is typically clear and free of visible particulate (unless the compound page states otherwise). Undissolved material is a reason to wait and swirl again, not to force through with a needle.
Common misconceptions
- Reconstitution volume is a choice, not a constant. Different diluent volumes for the same vial produce different concentrations. There is no single "correct" volume in the abstract; there is a volume the practitioner reference or protocol assumed.
- Reconstitution does not sterilise. If the diluent or technique was not sterile, the resulting solution is not sterile.
- "Fully dissolved" is not the same as "correctly reconstituted." A clear solution can still be at the wrong concentration if the volume was measured incorrectly.
Practical interpretation
Follow the compound page's diluent-volume convention when you want the Preparation Calculator's example values to line up with the underlying source. Use a different volume only when you understand what changes downstream.
